Physiological Perspectives on Molecular Mechanisms and Regulation of Vesicular Glutamate Transport: Lessons From Calyx of Held Synapses

SCAMP5 plays a critical role in axonal trafficking and synaptic localization of NHE6 to adjust quantal size at glutamatergic synapses

Summary: The study found that SCAMP5 interacts with NHE6 and regulates NHE6 recruitment to glutamatergic presynaptic terminals. Knockdown of SCAMP5 or perturbation of SCAMP5/NHE6 interaction inhibited axonal trafficking and the presynaptic localization of NHE6, leading to SV hyperacidification. This suggests that SCAMP5 plays a key role in regulating synaptic strength through adjustment of quantal size, and disruptions in their interaction may contribute to synaptic dysfunction in autism.

Chemical long-term potentiation increases the number of NHE6-positive presynaptic boutons, a process dependent on SCAMP5, without affecting the quantal size of glutamate release.
