2D Ti3C2TxMXene couples electrical stimulation to promote proliferation and neural differentiation of neural stem cells

Rongrong Guo et al.

Summary: Ti3C2Tx MXene nanosheets have shown the potential to regulate the fate of neural stem cells (NSCs) by promoting their proliferation and neural differentiation. NSCs cultured on Ti(3)C(2)T(x)MXene films exhibit enhanced electrical activity and more mature neurons with longer neurites and increased branching. Electrical stimulation applied through the Ti(3)C(2)T(x)MXene film further enhances NSC proliferation. This study expands the potential applications of MXene materials and provides new strategies for stem cell research.

Autophagy Regulates the Survival of Hair Cells and Spiral Ganglion Neurons in Cases of Noise, Ototoxic Drug, and Age-Induced Sensorineural Hearing Loss

Lingna Guo et al.

Summary: Inner ear hair cells and spiral ganglion neurons are crucial components of the auditory system, but they are susceptible to genetic defects, noise exposure, ototoxic drugs, and aging. Efforts to combat hearing loss have included cochlear implants, hair cell regeneration, gene therapy, and antioxidant drugs. Understanding the role of autophagy in sensorineural hearing loss and identifying potential autophagy-related targets could lead to new treatments for hearing loss.

Differences in Calcium Clearance at Inner Hair Cell Active Zones May Underlie the Difference in Susceptibility to Noise-Induced Cochlea Synaptopathy of C57BL/6J and CBA/CaJ Mice

Hongchao Liu et al.

Summary: This study found that moderate noise exposure can lead to permanent cochlear synaptopathy in CBA mice, while B6 mice fully recover. Molecularly, CBA mice showed enhanced Bax/Bcl-2 ratio and cytochrome-C expression after noise exposure, potentially disrupting calcium homeostasis.

Deletion of Kcnj16 in Mice Does Not Alter Auditory Function

Jun Lv et al.

Summary: The Kcnj16 gene that encodes Kir5.1 is not essential for auditory function in mice, as the loss of Kcnj16 did not significantly affect hair cell development or auditory function, despite a decrease in the expression of some ion channel proteins. The number and distribution of ribbon synapses also showed no significant differences between Kcnj16(-/-) mice and wild-type littermates.

FOXG1 promotes aging inner ear hair cell survival through activation of the autophagy pathway

Zu-Hong He et al.

Summary: Presbycusis, the result of aging on hearing, is closely related to common mitochondrial gene deletions. This study found that FOXG1 plays a crucial role in the degeneration process of the auditory system through regulation of autophagy. Aspirin was also found to increase FOXG1 expression and activate autophagy, suggesting a potential treatment for age-related hearing loss.

PRDX1 activates autophagy via the PTEN-AKT signaling pathway to protect against cisplatin-induced spiral ganglion neuron damage

Wenwen Liu et al.

Summary: Activated autophagic flux in SGNs after cisplatin treatment can alleviate SGN apoptosis and related hearing loss, indicating a neuroprotective role of autophagy. The antioxidant enzyme PRDX1 plays a crucial role in modulating autophagy in SGNs, which in turn activates the PTEN-AKT signaling pathway to trigger autophagy. These findings suggest potential therapeutic targets for drug-induced SNHL through autophagy activation.

Sonia Figuccia et al.

Summary: Mitochondrial protein synthesis is essential for oxidative phosphorylation, with mutations in the translation apparatus being a major cause of mitochondrial diseases; Aminoacyl-tRNA synthetases attach specific amino acids to tRNAs and are encoded by nuclear genes, in contrast to mttRNAs encoded by mitochondrial genes; Next-generation sequencing has identified an increasing number of mtARSs variants associated with clinical heterogeneity.

Mitochondrial Dysfunction and Sirtuins: Important Targets in Hearing Loss

Lingjun Zhang et al.

Summary: Mitochondrial dysfunction has been suggested as a risk factor for sensorineural hearing loss, mainly due to oxidative stress induced by reactive oxygen species (ROS) and mitochondrial DNA mutations. Mitochondrial sirtuins, particularly SIRT3, may modulate ROS levels in the pathology of hearing loss and potentially have a protective effect against it.

Deficiency of Klc2 Induces Low-Frequency Sensorineural Hearing Loss in C57BL/6 J Mice and Human

Xiaolong Fu et al.

Summary: Klc2 is essential for cochlear function, as its deficiency leads to abnormal transport of mitochondria and down-regulation of the GABAA receptor family, resulting in low-frequency hearing loss. WGS indicates KLC2's association with low-frequency hearing in humans, and delivering adeno-associated virus containing the Klc2 wide-type cDNA sequence can restore cochlear function.
