Developmental window of vulnerability to white matter injury driven by sublethal intermittent hypoxemia

Sergey A. Sosunov et al.

Summary: This study found that only neonatal mice exposed to early sublethal intermittent hypoxia (IH) showed arrested OL maturation, diffuse cerebral hypomyelination, and sensorimotor deficits, without any cellular death. This suggests that a developmental window of vulnerability to diffuse white matter injury (WMI) can exist even in the absence of OL lineage cellular death, indicating the involvement of cell-nonlethal mechanisms in defining this vulnerability.

Effects of nutrition therapy on growth, inflammation and metabolism in immature infants: a study protocol of a double-blind randomized controlled trial (ImNuT)

Kristina Wendel et al.

Summary: This study aims to investigate the effects of early supply of ARA and DHA on brain maturation and neonatal outcomes in infants born before 29 weeks of gestation. Infants will receive a fatty acid supplement from the second day of life to 36 weeks' postmenstrual age, with the primary outcome being brain maturation assessed by MRI.

Oxidative stress and impaired oligodendrocyte precursor cell differentiation in neurological disorders

Jan Spaas et al.

Summary: Oligodendrocyte precursor cells (OPCs) play a crucial role in various neurodegenerative disorders, with oxidative/carbonyl stress potentially impacting their differentiation and function, contributing to disease progression. Investigating how oxidative/carbonyl stress disrupts OPC differentiation may offer new directions for therapeutic strategies in neurodegenerative disorders.

Summary: Myelin, essential for efficient action potential propagation in the central nervous system, is now understood to be a dynamic process that can modulate neuronal function. Recent research has shown a reciprocal relationship between neurons and oligodendrocytes, highlighting the importance of their communication in refining neuronal function.

Identification of novel myelin repair drugs by modulation of oligodendroglial differentiation competence

Summary: The study identified novel small molecules that can promote oligodendroglial cell differentiation and found that some compounds can enhance human oligodendrogenesis and myelination in vitro. Steroid danazol and anthelminthic parbendazole were shown to increase myelin repair, suggesting their potential as therapeutic candidates for demyelinating diseases.

Early oxygen levels contribute to brain injury in extremely preterm infants

Krista Rantakari et al.

Summary: The study suggests that both abnormally low and high oxygen levels during early NICU care are harmful for later neurodevelopmental outcomes in preterm infants. Specific brain structures seem to be vulnerable to low oxygen levels, while others are vulnerable to high oxygen levels. Strict saturation targets during the early postnatal period in preterm infants are emphasized for clinical implications.

Myelin Repair: From Animal Models to Humans

Myriam Cayre et al.

Summary: It is possible for brain repair to occur through myelin regeneration, which involves various cell populations including oligodendrocyte progenitor cells. Factors such as aging, chronic inflammation, and extracellular matrix protein production can limit endogenous repair. Understanding the cells and mechanisms involved in myelin regeneration is crucial for developing new therapeutic strategies.

Myelination induces axonal hotspots of synaptic vesicle fusion that promote sheath growth

Rafael G. Almeida et al.

Summary: The research indicates that synaptic vesicle fusion plays a crucial role in the process of myelination, promoting the activity-regulated signal in neurons and consolidating sheath growth on specific axons. Axonal vesicular fusion increases significantly during myelination, impacting the formation and growth of myelin sheaths.

Pioglitazone Ameliorates Lipopolysaccharide-Induced Behavioral Impairment, Brain Inflammation, White Matter Injury and Mitochondrial Dysfunction in Neonatal Rats

Jiann-Horng Yeh et al.

Summary: The study demonstrated that pioglitazone can reduce behavioral deficits mediated by inflammatory-induced brain white matter injury in neonatal rats. Pioglitazone treatment improved LPS-induced neurobehavioral and physiological disturbances, possibly by attenuating LPS-induced increment of TBARS content, IL-1 beta levels and number of activated microglia. The neuroprotective effects of pioglitazone were associated with its impact on microglial activation, IL-1 beta induction, lipid peroxidation, oligodendrocyte production and mitochondrial activity.
