Summary: The aromatic ring in W-122 in the core domain of potyviral CP is critical for cell-to-cell movement by affecting CP stability and viral replication. Mutations in W-122 significantly reduce CP accumulation levels, impairing virus cell-to-cell movement. Substitution of W-122 with different aromatic residues allows mutant viruses to move systemically and accumulate to levels similar to wild-type viruses.

Double-stranded RNA confers resistance to pepper mottle virus in Nicotiana benthamiana

Summary: The study utilized RNAi technique to design dsRNA for protecting plants from viral infection, showing that optimizing dsRNA and virus injection time can enhance the protection efficiency. Different dsRNAs targeting various gene regions have varying effects on virus survival, with the highest inhibitory effect observed on the middle part of the NIb and HC-Pro genes. The results suggest that dsRNA tool can serve as a plant vaccine platform for crop protection.
