Stochastic Episodes of Latent Cytomegalovirus Transcription Drive CD8 T-Cell Memory Inflation and Avoid Immune Evasion

Marion Griessl et al.

Summary: This study investigated the transcription of viral genes during latent infection of murine cytomegalovirus, revealing that the transcription of antigenic peptides driving memory inflation rarely coincides with the transcription of immune evasion proteins. The stochastic transcription hypothesis explains why immune evasion does not interfere with memory inflation in latently infected cells.

Summary: HCMV infection is usually controlled by a vigorous immune response, but can cause serious organ diseases when the immune system is compromised. Significant progress has been made in understanding HCMV infection and disease, including defining the dynamics of viral replication and evaluating new antiviral drugs and vaccines.

Summary: Hematopoietic cell transplantation is a last resort for treating hematopoietic malignancies that are refractory to standard therapies, aiming to reconstitute hematopoiesis and immune function. Mismatches in HLA and minor histocompatibility antigens may increase the risk of complications from cytomegalovirus infection.

Direct Evidence for Viral Antigen Presentation during Latent Cytomegalovirus Infection

Niels A. W. Lemmermann et al.

Summary: Research on murine models of CMV infection has shown a phenomenon called memory inflation after primary CD8(+) T-cell response peaks. During CMV latency, there is an increase in cells specific for certain viral epitopes over time. Additionally, viral genes are not completely silent during latency and are expressed sporadically in a stochastic manner.

The m15 Locus of Murine Cytomegalovirus Modulates Natural Killer Cell Responses to Promote Dissemination to the Salivary Glands and Viral Shedding

Baca Chan et al.

Summary: The m15 locus of murine CMV plays a key role in modulating NK cell immunity, with disruption leading to attenuated dissemination and replication in the salivary glands. Disruption of the m15 locus is associated with heightened NK cell responses, and depletion of NK cells can rescue viral replication and shedding in the salivary glands. Multiple transcripts expressed by a single locus may work together to regulate the function of anti-viral NK cells.
