Efficacy and safety of temelimab in multiple sclerosis: Results of a randomized phase 2b and extension study

Hans-Peter Hartung et al.

Summary: The study did not meet its primary endpoint, but at 48 weeks participants treated with 18 mg/kg temelimab had fewer new T1-hypointense lesions. They also showed consistent, although statistically non-significant, reductions in brain atrophy and magnetization transfer ratio decrease compared to the placebo/comparator group. These trends were sustained over 96 weeks with no safety issues emerging.

Implication of human endogenous retrovirus W family envelope in hepatocellular carcinoma promotes MEK/ERK-mediated metastatic invasiveness and doxorubicin resistance

Yan Zhou et al.

Summary: Our study revealed that Syncytin-1 expression is up-regulated in HCC, particularly in advanced stages, and can serve as a predictor of patient prognosis. Furthermore, detailed analysis showed a significant association between Syncytin-1 and HBsAg positivity in HCC patients, suggesting a key role in the development and treatment of HCC.

Human endogenous retrovirus W family envelope protein (HERV-W env) facilitates the production of TNF-α and IL-10 by inhibiting MyD88s in glial cells

Xiuling Wang et al.

Summary: The study revealed that HERV-W env upregulates the expressions of TNF-alpha and IL-10 by inhibiting the production of MyD88s in glial cells. This immune pathogenesis mechanism involving TLR4 and MyD88 sheds light on the role of HERV-W env in neuropsychiatric disorders.

Phagocyte-mediated synapse removal in cortical neuroinflammation is promoted by local calcium accumulation

Mehrnoosh Jafari et al.

Summary: Synapse loss in the cortex is a key pathological feature in multiple sclerosis, which contributes to chronic cognitive impairment. Gray matter inflammation affects neuronal structure and function, and targeting synapse loss through immunomodulatory therapy could be a potential treatment strategy.

CPEB1, a novel risk gene in recent-onset schizophrenia, contributes to mitochondrial complex I defect caused by a defective provirus ERVWE1

Ya-Ru Xia et al.

Summary: CPEB1 and NDUFV2 are potential blood-based biomarkers and pathogenic factors in schizophrenia. The study reveals a novel mechanism of ERVWE1 in the etiology of schizophrenia, showing its role in inhibiting complex I activity in SH-SY5Y cells through the CPEB1/NDUFV2P1/NDUFV2 signaling pathway.

Identification of inflammatory subgroups of schizophrenia and bipolar disorder patients with HERV-W ENV antigenemia by unsupervised cluster analysis

Ryad Tamouza et al.

Summary: Our study found that subsets of patients with schizophrenia (approximately 41%) and bipolar disorder (approximately 28%) show positive antigenemia for HERV-W ENV protein, while the large majority of healthy controls (96%) was found to be negative. Unsupervised cluster analysis identified two main clusters of patients, best predicted by the presence or absence of HERV-W ENV protein. HERV-W expression was associated with increased serum levels of inflammatory cytokines and higher childhood maltreatment scores.
