Microbiota profiling and screening of the lipase active halotolerant yeasts of the olive brine

Summary: In this study, microbial analysis of cracked green olive brine from southern Turkey was conducted for the first time, revealing a high percentage of fungi, particularly from the phylum Ascomycota. Yeasts grown in high salt concentrations were found to produce lipase with high similarity to Candida diddensiae, with subsequent analysis showing cell-bound lipase activity at pH 3.0.

Summary: The study focused on the successful degradation of high concentrations of polyaromatic phenanthrene and benzo[a]pyrene by the extremotolerant yeast Rhodotorula mucilaginosa from Arctic sea ice under hypersaline conditions. The results showed a high efficiency in PAH degradation and non-toxic metabolites production. Transcriptome analysis revealed differences in gene expression between PAHs and glucose, while genomic mining indicated a widespread distribution of xenobiotic degradation pathways in different yeast lineages.

The revenge of Zygosaccharomyces yeasts in food biotechnology and applied microbiology

Summary: Non-conventional yeasts like Zygosaccharomyces rouxii and Zygosaccharomyces bailii are known for spoiling food and beverages, but their ability to adapt to various process conditions make them attractive for bio-conversion. Z. rouxii plays a key role in converting sugars in balsamic vinegar and releasing important flavor compounds in soy sauce, while Z. bailii shows promise in lactic acid and bioethanol production. Research efforts focusing on genetic bases of these yeasts' distinctive traits are expected to enhance their applications in biotechnology and applied microbiology.

Biodegradation of phenol by a halotolerant versatile yeast Candida tropicalis SDP-1 in wastewater and soil under high salinity conditions

Summary: This study isolated a novel halotolerant phenol-degrading yeast strain, SDP-1, from a coastal soil in Jiangsu, China, and identified it as Candida tropicalis based on morphology and genetic sequence analysis. SDP-1 efficiently removes phenol under a wide range of conditions, utilizes various phenol derivatives as carbon sources, and shows high tolerance to salts. Immobilized SDP-1 exhibits enhanced phenol degradation efficiency compared to free cells, making it a potential candidate for phenol-contaminated wastewater and soil treatment in hypersaline environments.

A CRISPR/Cas9 method facilitates efficient oligo-mediated gene editing in Debaryomyces hansenii

Summary: The study introduces a novel plasmid-based CRISPR-CUG/Cas9 method for easy and efficient gene editing in prototrophic strains of D. hansenii, enabling multiplex gene engineering. Additionally, NHEJ-deficient D. hansenii strains have been constructed to support highly efficient introduction of point mutations and single/double gene deletions. Furthermore, the use of 90-nt single-stranded DNA oligonucleotides for direct repair of DNA breaks induced by sgRNA-Cas9 resulted in precise edits with 100% efficiency.
