Modulating effects of RAMPs on signaling profiles of the glucagon receptor family

Summary: This study comprehensively investigated the interactions between receptor activity-modulating proteins (RAMPs) and G protein-coupled receptors (GPCRs) using a bioluminescence resonance energy transfer (BRET) approach. The results revealed that RAMPs modulate the signal transduction of glucagon receptor family members in a receptor-specific manner. These findings provide new insights into the role of RAMPs in ligand recognition and receptor activation.

Although phosphorylation is a major regulator of GPCR signaling, other post-translational modifications (PTMs) such as ubiquitination, glycosylation, and palmitoylation play important roles in GPCR biology, yet understanding of these PTMs is limited. Comprehensive understanding of diverse PTMs is crucial for elucidating dysregulated mechanisms in disease and improving drug development for GPCRs.

This new mechanism of sustained signaling presents new research directions and challenges.

Constitutive signal bias mediated by the human GHRHR splice variant 1

Summary: The study found that a splice variant (SV1) of the human growth hormone-releasing hormone receptor is capable of transducing biased signal, selectively coupling to beta-arrestins while the main receptor activates Gs. Differences at the receptor N terminus lead to distinct downstream signaling pathways for GHRHR and SV1, with GHRHR predominantly activating Gs and SV1 selectively coupling to beta-arrestins.
