Protein Modification Characteristics of the Malaria Parasite Plasmodium falciparum and the Infected Erythrocytes

Jianhua Wang et al.

Summary: Extensive and dynamic posttranslational modifications of proteins in Plasmodium falciparum and infected red blood cells play a crucial role in understanding parasite biology and malaria pathogenesis. These modifications critically modulate virulence factors, host-parasite interaction, and disease mechanisms in malaria. Furthermore, the study provides a valuable resource for mining new antimalarial targets.

20S proteasomes secreted by the malaria parasite promote its growth

Elya Dekel et al.

Summary: The study demonstrates that Plasmodium falciparum secretes extracellular vesicles (EVs) containing functional 20S proteasome complexes which remodel the cytoskeleton of naive human red blood cells, preparing them for parasite invasion.

Plasmodium vivax infection compromises reticulocyte stability

Martha A. Clark et al.

Summary: During Plasmodium intra-erythrocytic development, parasites compromise the structural integrity of host red-blood cells. Researchers found that P. vivax infection destabilizes host reticulocytes, which are less stable than normocytes infected with P. falciparum.

The plasma membrane calcium ATPase 4 does not influence parasite levels but partially promotes experimental cerebral malaria during murine blood stage malaria

Ana Villegas-Mendez et al.

Summary: Recent genome wide analysis studies have found a strong association between genetic variations in the human ATP2B4 gene and susceptibility to severe malaria, suggesting that the plasma membrane calcium ATPase 4 (PMCA4) encoded by this gene may play a role in modulating the growth and invasion of Plasmodium parasite within red blood cells. Knockout of PMCA4 in mice did not significantly affect parasite levels in different Plasmodium infections, but did provide slight protection against experimental cerebral malaria, possibly through a reduction in brain T cell accumulation.

Illuminating Plasmodium invasion by lattice-light-sheet microscopy

Summary: Plasmodium merozoites invade erythrocytes through ligand binding, calcium signaling, and membrane deformation in a stepwise manner. Geoghegan et al. utilized a newly developed light sheet microscope to investigate invasion with unprecedented temporal resolution, revealing the roles of host cell cholesterol and pore formation at the parasite-host cell interface.

Summary: Host membrane remodeling plays a crucial role in the invasion process of Plasmodium parasites by predominantly forming the parasitophorous vacuole from the erythrocyte membrane and continuously remodeling it throughout invasion.
