New frontiers in translational research: Touchscreens, open science, and the mouse translational research accelerator platform

Jacqueline A. Sullivan et al.

Summary: Neurodegenerative and neuropsychiatric diseases often lead to impairments in high-level cognitive functions, complicating the search for new treatments. The Mouse Translational Research Accelerator Platform (MouseTRAP) integrates touchscreen cognitive testing and neurotechnology to facilitate translational research, addressing key obstacles such as the lack of robust cognitive assessment tools. Touchscreencognition.org and mousebytes.ca are Open Science platforms designed to share knowledge and data within the rodent touchscreen community, enhancing collaboration and improving research outcomes.

Loss of α7 nicotinic acetylcholine receptors in GABAergic neurons causes sex-dependent decreases in radial glia-like cell quantity and impairments in cognitive and social behavior

Samir A. Nacer et al.

Summary: The dentate gyrus is a unique brain structure where neurons can be generated postnatally and integrated into existing circuitry throughout life. Nicotinic acetylcholine receptors (nAChRs), particularly the alpha 7 subtype, play a crucial role in modulating the maturation process of newly generated neurons. Knocking out alpha 7 nAChRs in GABAergic neurons results in reduced radial glia-like cells, decreased immature neurons, and sex-dependent changes in behavior, including social recognition and spatial learning. These findings highlight the importance of alpha 7 nAChRs in regulating adult neural stem cell pool and behavior.

Summary: This study reveals the molecular mechanism behind the subnucleus-specific alterations in BFCN excitability caused by the interaction between oAβ(42) and nAChR receptors, as well as confirms the important role of α7β2-nAChR in oAβ(42)-induced cognitive decline.

Kasey P. Spry et al.

Summary: Object recognition tasks are commonly used assays for studying learning and memory in rodents, but inconsistencies in conducting these tasks across labs exist. By designing and using standardized 3D-printed objects and utilizing capacitive sensing technology to measure object investigation, these inconsistencies can be resolved.

Summary: Cigarette smoking is a major risk factor for cardiovascular diseases, but the role of nicotine in the development of CVD is not fully understood. Public awareness of smoking dangers has reduced smoking prevalence, but the use of e-cigarettes has increased due to perceived safety. This review discusses the effects of nicotine on the vasculature and calls for further research on the topic.

Summary: The systematic review and meta-analysis of 31 publications involving 978 subjects found that transdermal nicotine administration has significant positive effects on attention, but the effects on memory were non-significant in healthy non-smoking adults. Further studies are encouraged to explore the therapeutic potential of nicotine patches in cognitive disorders.

Structure and gating mechanism of the α7 nicotinic acetylcholine receptor

Colleen M. Noviello et al.

Summary: The alpha 7 nicotinic acetylcholine receptor plays critical roles in the central nervous system and the cholinergic inflammatory pathway as a ligand-gated ion channel. Recent cryo-EM structures of the human alpha 7 nicotinic receptor in different states have provided insights into its gating cycle and functional elements, including a C-terminal latch and an anionic ring that contribute to its high conductance and calcium permeability. Comparisons among the alpha 7 structures have revealed divergence in gating mechanisms within the Cys-loop receptor superfamily.

Differential signalling induced by α7 nicotinic acetylcholine receptors in hippocampal dentate gyrus in vitro and in vivo

Qing Cheng et al.

Summary: The study compared the responses of dentate granule cells and GABAergic interneurons to α7 nAChR agonists, revealing differential effects on intracellular signaling pathways possibly due to variations in Ca2+ regulation in different cell types. Additionally, in vivo observations showed enhanced neuronal activity in dentate granule cells but not in GABAergic neurons after nicotine administration, indicating a cell type-specific mechanism for α7 nAChR-mediated increase of neuronal activity.

Sleep loss drives acetylcholine- and somatostatin interneuron-mediated gating of hippocampal activity to inhibit memory consolidation

James Delorme et al.

Summary: Sleep loss disrupts the consolidation of hippocampus-dependent memory by activating hippocampal somatostatin-expressing interneurons and cholinergic and orexinergic hippocampal inputs. The activation or inhibition of these cell populations can gate activity in granule cells and affect memory consolidation. Additional features of sleeping brain activity are also required for memory consolidation, suggesting a complex mechanism involving cholinergic input and local Sst+ interneurons in mediating state-dependent gating of DG activity.
