Identification of novel risk loci with shared effects on alcoholism, heroin, and methamphetamine dependence

Yan Sun et al.

Summary: This study identified and replicated three novel genetic loci associated with the common risk of heroin, methamphetamine addiction, and alcoholism. These loci have functional effects on gene expression and brain structure, and may provide insights into the root cause of substance dependence, aiding in the development of prevention strategies for addiction disorders.

Differential correlation of serum BDNF and microRNA content in rats with rapid or late onset of heavy alcohol use

Yann Ehinger et al.

Summary: Heavy alcohol use reduces BDNF levels in the prefrontal cortex of rodents through upregulation of miRs targeting BDNF mRNA. In humans, there is an inverse correlation between circulating BDNF levels and psychiatric disorders severity. This study found that rats exhibit different drinking patterns during heavy alcohol use, with serum BDNF levels negatively correlated with alcohol intake.

Kappa opioid receptor and dynorphin signaling in the central amygdala regulates alcohol intake

Daniel W. Bloodgood et al.

Summary: Excessive alcohol consumption is influenced by the signaling of preprodynorphin and Kappa Opioid Receptor in the central nucleus of the amygdala (CeA), with conditional knockout of these genes resulting in sex-specific responses. Knockout of PDYN gene decreased alcohol drinking in both male and female mice, while knockout of KOR gene decreased drinking in males only.

Differential regulation of alcohol consumption and reward by the transcriptional cofactor LMO4

Rajani Maiya et al.

Summary: Repeated alcohol exposure leads to changes in gene expression that may contribute to alcohol dependence, with LMO4 identified as a potential regulator in this process. Knockdown of Lmo4 in the nucleus accumbens increases alcohol consumption, while knockdown in the basolateral amygdala decreases alcohol consumption and preference. Transcriptome profiling reveals different targets of LMO4 in these brain regions, indicating a complex regulatory network underlying alcohol consumption.

cAMP-Fyn signaling in the dorsomedial striatum direct pathway drives excessive alcohol use

Yann Ehinger et al.

Summary: The Fyn kinase in the dorsomedial striatum of rodents has been found to play a crucial role in mechanisms underlying excessive alcohol intake, with its actions being cell-type specific to dMSNs. The activation of cAMP-Fyn axis in DMS dMSNs is identified as a molecular transducer of mechanisms leading to the development of excessive alcohol consumption.

Compulsive alcohol consumption is regulated by dorsal striatum fast-spiking interneurons

Michael S. Patton et al.

Summary: These findings suggest that the fast-spiking interneurons in the striatum play a crucial role in driving compulsive ethanol consumption. Ablation of these neurons reduces ethanol consumption and eliminates compulsive drinking behavior, highlighting the importance of this particular neural population in addictive behaviors.

Role of FMRP in rapid antidepressant effects and synapse regulation

Chelcie F. Heaney et al.

Summary: Rapid antidepressants work by regulating FMRP expression in dendrites and modulating the mTORC1 pathway. The behavioral effects of Ro-25-6981 are dependent on FMRP expression and its differential mRNA binding controlled by mTORC1. In animal models with FXS, GABA(B)R activity can hinder the effects of Ro-25-6981, indicating a need for combined therapy targeting GABA(B)Rs and NMDARs in individuals with comorbid FXS and MDD.

Long-term alcohol consumption alters dorsal striatal dopamine release and regulation by D2 dopamine receptors in rhesus macaques

Armando G. Salinas et al.

Summary: The study found that chronic alcohol consumption can lead to adaptations in the dopamine system, with sex-dependent alterations in dopamine release in different subregions of the dorsal striatum. Male rhesus macaques showed reduced dopamine release in the putamen following alcohol consumption, while only male macaques in abstinence had reduced dopamine release in the caudate. In contrast, female macaques showed enhanced dopamine release in the caudate but not in the putamen.

Drug Addiction: Hyperkatifeia/Negative Reinforcement as a Framework for Medications Development

George F. Koob

Summary: Compulsive drug seeking behavior associated with addiction may be driven by a heuristic framework involving three stages and three domains of dysfunction, with hyperkatifeia playing a role as a source of motivation through negative reinforcement. Neurochemical/neurocircuitry dysregulations could mediate the occurrence of a negative hedonic set point, ultimately leading to compulsive drug seeking behavior.

Alcohol use disorder causes global changes in splicing in the human brain

Binge-like ethanol drinking activates anaplastic lymphoma kinase signaling and increases the expression of STAT3 target genes in the mouse hippocampus and prefrontal cortex

Kana Hamada et al.

Summary: This study identified activation of the ALK-STAT3 signaling pathway in the brain after binge-like ethanol consumption, pinpointed potential novel ethanol-responsive STAT3 target genes, and suggested that inhibition of STAT3 could be a potential method to reduce binge drinking in humans.

A Circuit-Based Information Approach to Substance Abuse Research

David M. Lovinger et al.

Summary: Research on substance-use disorders (SUDs) in animals has focused on learning models and identifying 'addiction-prone' animals, while basic neuroscientific research has advanced in understanding molecular, cellular, and circuit functions. However, debates over addiction-like animal models and the correlation between behaviors and brain regions hinder SUD-related research. A 'bottom-up' approach involving characterization of brain circuits and understanding substance use patterns and neurobiological effects can benefit SUD-related research.

Brain-specific inhibition of mTORC1 eliminates side effects resulting from mTORC1 blockade in the periphery and reduces alcohol intake in mice

Yann Ehinger et al.

Summary: Alcohol Use Disorder (AUD) is a common issue with limited treatment options. Research suggests mTORC1 as a potential therapeutic target, but chronic use may lead to severe side effects. The study proposes a binary drug strategy, utilizing a combination of drugs to protect mTORC1 activity in the periphery and mitigate adverse effects.

A neural substrate of compulsive alcohol use

Esi Domi et al.

Summary: Research using a footshock-punished alcohol self-administration procedure identified a subpopulation of rats showing punishment-resistant self-administration behavior, which was found to be a stable trait in a subset of rats and associated with activity in the CeA brain network. Activity of PKC delta(+) inhibitory neurons in the CeL accounted for a significant portion of variance in punishment-resistant alcohol taking, suggesting a potential therapeutic target for alcohol addiction.
