Insulin-promoted mobilization of GLUT4 from a perinuclear storage site requires RAB10

Alexandria Brumfield et al.

Summary: The TBC1D4-RAB10 signaling module is found to control GLUT4 mobilization from the TGN storage compartment, indicating that insulin regulates GLUT4 behavior not only at the plasma membrane but also deeper within the cell. Results show that the GLUT4-containing region of the TGN functions as a sorting and storage site, where various cargoes are mobilized by distinct signals through unique molecular machinery.

Insulin action in adipocytes, adipose remodeling, and systemic effects

Anna Santoro et al.

Summary: On the 100th anniversary of the discovery of insulin, the critical role of adipocytes in determining insulin action at the cellular level is recognized. Adipose tissue biology has greatly evolved, revealing that adipocytes are more than just storage depots for fat. Adipocytes, in response to insulin, play an important role in controlling whole-body nutrient homeostasis through their plasticity, heterogeneity, and endocrine function.

The many actions of insulin in skeletal muscle, the paramount tissue determining glycemia

Lykke Sylow et al.

Summary: Skeletal muscle is crucial for insulin-stimulated glucose disposal and overall glycemic control. Insulin regulates glucose uptake in skeletal muscle through complex signaling cascades. Understanding insulin's actions in muscle can provide insights into addressing issues with glucose uptake in conditions like diabetes and obesity.
