Preclinical assessment of the efficacy and specificity of GD2-B7H3 SynNotch CAR-T in metastatic neuroblastoma

Babak Moghimi et al.

Summary: The study highlights the importance of developing gated systems for predicting clinical toxicity of CAR T cells in solid tumors. By utilizing murine and human CAR-T cells specific for tumor antigens, the researchers were able to generate a GD2-B7H3 CAR-T with improved metabolic fitness and efficacy in controlling neuroblastoma growth in preclinical models. This innovative approach shows promising results in suppressing neuroblastoma with minimal adverse effects.

DutaFabs are engineered therapeutic Fab fragments that can bind two targets simultaneously

Roland Beckmann et al.

Summary: The study introduces a platform of dual targeting Fab molecules with two independent binding sites, allowing simultaneous binding of two target molecules. These molecules exhibit high affinity, physico-chemical stability, solubility, and superior efficacy compared to anti-VEGF monotherapy in vivo.

Active-site loop variations adjust activity and selectivity of the cumene dioxygenase

Peter M. Heinemann et al.

Summary: Active-site loops are crucial for enzyme catalytic properties, but challenging to engineer due to their high flexibility and diversity. The authors identify and engineer hot-spots in the loops of cumene dioxygenase, obtaining variants with altered activity, regio- and enantioselectivity, and present a valuable Linker In Loop Insertion approach for enhanced biocatalysts in the future.

A single-chain antibody generation system yielding CAR-T cells with superior antitumor function

Toshiki Ochi et al.

Summary: Ochi et al. have established a system to generate and screen a library of CAR-T cells expressing different immunoglobulin variable regions, which can enhance the anti-tumour activities of CAR-T cells in vivo. By optimizing the variable region of CAR-T cells, target-specific recognition can be fine-tuned, resulting in better proliferation capacity and durable phenotypes against advanced tumors.

Chimeric STAR receptors using TCR machinery mediate robust responses against solid tumors

Yue Liu et al.

Summary: A new double-chain chimeric receptor STAR was constructed to mediate strong and sensitive TCR-like signaling upon antigen stimulation, showing better antigen sensitivity and less risk of dysfunction compared to traditional CAR-T cells. In multiple solid tumor models, STAR-T cells outperformed BBzCAR-T cells and showed superior antitumor effects.

An engineered ligand trap inhibits leukemia inhibitory factor as pancreatic cancer treatment strategy

Sean A. Hunter et al.

Summary: Hunter et al. engineered a high affinity, soluble variant of leukemia inhibitory factor receptor (LIFR) to serve as a ligand trap for the LIF cytokine. They demonstrated that this engineered LIFR exhibits improved affinity relative to the wild-type receptor, leading to better disruption of LIF signaling in cancer cells, and highlighting promise of such ligand traps as therapeutic strategy for cancer treatment.

Durable Responses and Low Toxicity After Fast Off-Rate CD19 Chimeric Antigen Receptor-T Therapy in Adults With Relapsed or Refractory B-Cell Acute Lymphoblastic Leukemia

Claire Roddie et al.

Summary: A novel second-generation CD19-CAR therapy (AUT01) has shown promising results in the treatment of relapsed or refractory adult B-cell acute lymphoblastic leukemia (B-ALL), demonstrating tolerable safety profile, high remission rates, and excellent persistence. This therapy has the potential to become a stand-alone treatment option for r/r adult B-ALL.
