Role of sphingolipid metabolism in neurodegeneration

Summary: Sphingolipid metabolism plays a crucial role in neurodegenerative diseases, affecting neuron development, health, and inducing neurotoxicity in the CNS. Gangliosides and sphingosine-1-phosphate contribute significantly to the pathogenesis of neurologic diseases and hold potential as therapeutic targets for the modulation of neurodegeneration.

Toward a Better Testing Paradigm for Developmental Neurotoxicity: OECD Efforts and Regulatory Considerations

Summary: Recognizing the current DNT testing paradigm is not suitable for assessing a large number of chemicals, scientific advances have been made in alternative methods to evaluate chemical interactions with the developing nervous system without using animals. The OECD provides internationally harmonised guidelines based on cellular models for testing and assessing chemicals for DNT. To address challenging endpoints like DNT, the OECD has been developing integrated approaches for testing and assessment, which rely on a combination of different data layers and mechanistic knowledge.

Changes in Sphingolipid Profile of Benzo[a]pyrene-Transformed Human Bronchial Epithelial Cells Are Reflected in the Altered Composition of Sphingolipids in Their Exosomes

Summary: The study identified significant alterations in the SL and GSL profile in transformed cells and their exosomes, with a majority of SLs and GSLs transported by exosomes in a similar relative pattern as in the cells of origin. The exceptions included decreased contents of certain substances and increased levels of others. These changes might contribute to cancer progression in recipient cells.
