Neutralizing Activity of Broadly Neutralizing Anti-HIV-1 Antibodies against Primary African Isolates

Julio C. C. Lorenzi et al.

Summary: This study examined the neutralizing activity of 9 anti-HIV-1 bNAbs against primary African isolates, finding that the potency and breadth of bNAbs were significantly lower than what was seen with pseudotyped virus panels. The differences in sensitivity between pseudotyped viruses and primary isolates varied depending on the bNAb and the HIV-1 Glade.

Recapitulation of HIV-1 Env-antibody coevolution in macaques leading to neutralization breadth

Ryan S. Roark et al.

Summary: Research has shown that in rhesus macaques, primary HIV-1 envelope proteins can trigger patterns of coevolution with antibodies similar to those observed in humans, providing precise solutions for virus persistence. These findings can guide the design of HIV-1 vaccines by recapitulating the developmental features of human broadly neutralizing antibodies.

Affinity maturation of SARS-CoV-2 neutralizing antibodies confers potency, breadth, and resilience to viral escape mutations

Frauke Muecksch et al.

Summary: This study examined the development of antibodies following infection with the coronavirus, finding that evolved antibodies had increased affinity and neutralization potency, altered mutational pathways for viral resistance, and restricted neutralization escape options. These findings suggest that increasing antibody diversity through prolonged or repeated antigen exposure may improve protection against diversifying populations of the virus and other pandemic threat coronaviruses.

Disassembly of HIV envelope glycoprotein trimer immunogens is driven by antibodies elicited via immunization

Hannah L. Turner et al.

Summary: The study found that off-target antibodies elicited by HIV trimer subunit vaccines can cause protein degradation into protomers, exposing more off-target epitopes and leading to further off-target antibody responses. This provides critical insights for improving HIV subunit trimer vaccines in future iterations of the vaccine design process.
