Summary: High-throughput sequencing is commonly used for studying soil microbial communities, but the choice of primers targeting different regions of 16S rRNA can significantly impact the diversity and structure of microbial communities. This study found that the primer pairs 338f/806r and 520f/802r produced different results in assessing bacterial communities in soils of varying land uses. While primer pair 520f/802r showed no significant impact of land use on soil bacterial diversity, primer pair 338f/806r revealed distinct variations. Additionally, the soil bacterial diversity generated by primer pair 338f/806r was influenced by environmental factors, while primer pair 520f/802r showed no significant correlations.
