Bringing Themes: Short Protein Segments Found in Different Architectures

Rachel Kolodny et al.

Summary: The majority of polypeptide chains cannot fold, making protein evolution from preexisting building blocks advantageous. Sequence similarities between different proteins suggest common ancestry, and short themes shared between domains may provide clues about domain emergence.

Searching protein space for ancient sub-domain segments

Rachel Kolodny

Summary: Evolutionary processes have left traces of reused homologous segments, or 'themes,' in the current protein universe, with recent studies focusing on sub-domain-scale themes and their potential insights into more ancient events. Particularly interesting are themes that are reused across dissimilar contexts, providing clues to the emergence of protein domains. Computational studies have identified such reused themes within different contexts at the sub-domain level.

Repeat proteins: designing new shapes and functions for solenoid folds

Frances Gidley et al.

Summary: The modular nature of repeat proteins has led to the design of novel sequences and structures with a broad set of design approaches. These proteins have found applications in molecular recognition by taking advantage of their natural ability to bind proteins, peptides and nucleic acids. New architectures combining different types of repeat can be easily scalable by changing the number of repeats and quickly generated using existing modular building blocks.

A small-molecule inhibitor of the BRCA2-RAD51 interaction modulates RAD51 assembly and potentiates DNA damage-induced cell death

Duncan E. Scott et al.

Summary: The study identifies CAM833 as an orthosteric inhibitor of RAD51:BRC, which affects DNA repair and potentiates cell death induced by DNA damage, working in coordination with PARP1 inhibitors to suppress growth in BRCA2-wildtype cells.
