The upregulation of PYCR2 is associated with aggressive colon cancer progression and a poor prognosis

Sitong Wang et al.

Summary: PYCR2 has been shown to be upregulated in various cancer types, correlating with poor patient prognosis and malignant clinicopathological characteristics. It is identified as an independent predictor of survival in colon cancer patients, and knocking down PYCR2 disrupts the activities of colon cancer cells. Gene set enrichment analysis suggests high PYCR2 expression correlates with the enrichment of specific signaling pathways.

Regulation of Extracellular Matrix Production in Activated Fibroblasts: Roles of Amino Acid Metabolism in Collagen Synthesis

Emily J. Kay et al.

Summary: This review summarizes the role of cancer associated fibroblasts (CAFs) in regulating ECM production, with a focus on collagen production, at the transcriptional, translational, and post-translational levels, and discusses possible strategies for effectively targeting CAF activation and the formation of a tumor-promoting stroma.

Mitochondrial NADP+ is essential for proline biosynthesis during cell growth

Diem H. Tran et al.

Summary: Research has shown that mitochondrial NADPH is crucial for proline biosynthesis in cells, with NAD kinase 2 (NADK2) activity being necessary for the production of mitochondrial NADP(+). Deficiency in mitochondrial NADPH leads to the inability of cells to synthesize proline, impacting nucleotide and protein synthesis, thereby impeding cell growth.
