RNA structure probing reveals the structural basis of Dicer binding and cleavage

Summary: Sequencing methods like icSHAPE have been developed to probe RNA structures transcriptome-wide in cells. The authors of this study introduced icSHAPE-MaP, a method combining in vivo selective 2'-hydroxyl acylation and mutational profiling to probe intact RNA structures, revealing distinct structural categories of Dicer substrates in correlation to both their binding affinity and cleavage efficiency. By analyzing RNA structural data using tertiary structural modeling constrained by icSHAPE-MaP, the study found that spatial distance measuring is an influential parameter for Dicer cleavage-site selection.
