Probing biased activation of mu-opioid receptor by the biased agonist PZM21 using all atom molecular dynamics simulation

Siyan Liao et al.

Summary: This study compared the binding and action mechanisms of the MOR biased agonist PZM21 and morphine to the mu-opioid receptor, revealing that PZM21 preferentially activates the G protein pathway over the beta-arrestin pathway. The interaction between PZM21 and MOR led to conformational changes and altered amino acid interactions within the receptor, potentially facilitating G protein binding.

Delineating the conformational landscape of the adenosine A2A receptor during G protein coupling

Shuya Kate Huang et al.

Summary: This study used fluorine nuclear magnetic resonance spectroscopy to identify key functional states of the adenosine A(2A) receptor complexed with heterotrimeric G protein in a phospholipid membrane environment. The findings revealed an ensemble consisting of inactive states, a G-protein-bound activation intermediate, and nucleotide-free states associated with agonist-driven activation. The crucial role of the G beta gamma subunit in facilitating ligand-dependent allosteric transmission was demonstrated.

Revealing atomic-scale molecular diffusion of a plant-transcription factor WRKY domain protein along DNA

Liqiang Dai et al.

Summary: This study utilized molecular dynamics simulations to reveal the diffusion dynamics of plant WRKY domain protein along DNA, uncovering its preferential binding to one DNA strand. The study also captured a complete one-base-pair stepping cycle of the protein on DNA, as well as tracking motions along the major groove and occasional crossing to the minor groove. The findings provide insights into how small TF domain proteins approach specific recognition sites on DNA and can support high-precision experimental detection.
