However, around 50% of cases remain without a known etiology, suggesting a strong genetic component. Recurrent KCNT2 missense variants affecting p.Arg190 result in a recognizable phenotype

Summary: Two individuals with intellectual disability, dysmorphic features, hypertrichosis, macrocephaly, and de novo KCNT2 missense variants affecting the Arg190 residue are reported. These variants are likely to disrupt the stabilization of a closed channel conformation of KCNT2, leading to a constitutively open state. This is the first report of KCNT2 pathogenic variants causing a developmental phenotype without epilepsy.
