Selective Synthesis of Pyrano[3,2-b]indoles or Cyclopenta[b]indoles Tethered with Medium-Sized Rings via Cascade C-C σ-Bond Cleavage and C-H Functionalization

Summary: Highly atom-economical tandem reactions have been developed for the synthesis of pyrano[3,2-b]indoles or cyclopenta[b]indoles tethered with 7-, 8-, or 9-membered rings. These reactions involve a carbon-carbon sigma-bond cleavage reaction of cyclic beta-ketoesters followed by intramolecular O-H/C-H coupling to give pyrano[3,2-b]indoles or C3 nucleophilic addition to afford cyclopenta[b]indoles.

A Thiol-Mediated Three-Step Ring Expansion Cascade for the Conversion of Indoles into Functionalized Quinolines

Summary: An operationally simple, high yielding three-step cascade process has been developed for the direct conversion of indole-tethered ynones into functionalized quinolines. A single multitasking thiol reagent is used to promote a three-step dearomatizing spirocyclization, nucleophilic substitution, and one-atom ring expansion reaction cascade under remarkably mild conditions. Additionally, a novel route to thio-oxindoles was discovered serendipitously.

Carbon Atom Insertion into Pyrroles and Indoles Promoted by Chlorodiazirines

Summary: This study presents a reaction for selectively generating 3-arylpyridine and quinoline motifs by inserting aryl carbynyl cation equivalents into pyrrole and indole cores. The use of α-chlorodiazirines as thermal precursors allows for a modification of the traditional protocol to directly afford these compounds. Selectivity based on pyrrole substitution patterns is examined, with a predictive model proposed based on steric effects.
