HDAC3 Activity within the Nucleus Accumbens Regulates Cocaine-Induced Plasticity and Behavior in a Cell-Type-Specific Manner

Summary: This study explores the impact of cocaine on HDAC3-dependent mechanisms, finding that chronic cocaine alters promoter-specific changes in HDAC3 and H4K8Ac occupancy, correlated with changes in expression of plasticity-related genes. Disrupting HDAC3's enzymatic activity changes gene expression and synaptic plasticity, without affecting cocaine-induced behaviors. Additionally, chronic cocaine increases Hdac3 expression in Drd1 cells, suggesting a cell-type-specific role of HDAC3 in regulating cocaine-associated memory formation and cocaine-seeking behavior.

Epigenetic Landscape of Methamphetamine Use Disorder

Summary: This review discusses the evidence of methamphetamine exposure causing changes in epigenetic modifications and the correlations between these changes and behavioral alterations. Studies on rats have shown associations between prolonged methamphetamine self-administration with compulsive drug-taking and abstinent phenotypes, as well as increased DNA hydroxymethylation and mRNA expression in genes encoding potassium channels in the nucleus accumbens. Further research is needed to understand how pharmacological or genetic manipulations of epigenetic alterations and potassium channel expression can affect methamphetamine-taking behaviors and relapse after long periods of abstinence.
