MALDI-TOF-Based Affinity Selection Mass Spectrometry for Automated Screening of Protein-Ligand Interactions at High Throughput

Roman P. Simon et al.

Summary: This study introduces a ligand binding assay platform that combines automated affinity selection (AS) sample preparation and label-free matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) analysis, allowing for mass spectrometry-based high-throughput screening of small-molecule target interactions. The platform is capable of identifying orthosteric and allosteric ligands, enabling efficient separation of target-ligand comp

Fast Nanoliter-Scale Cell Assays Using Droplet Microarray-Mass Spectrometry Imaging

Carina RamalloGuevara et al.

Summary: In pharmaceutical research and development, cell-based assays utilize fluorescence and label-dependent techniques for analyzing various cellular processes. Recently, superhydrophobic-hydrophilic droplet microarrays (DMA) and matrix-assisted laser desorption/ionization (MALDI) mass spectrometry have emerged as key technologies for miniaturized high-throughput cell assays and label-free molecular high-content drug profiling. The integration of nanoliter-scale cell assays on DMAs with MALDI-MS imaging approaches on a DMA-MSI platform has shown promise in concentration-response profiling of pharmaceutical compounds and allows for MALDI imaging analysis of as few as ten cells per spot through miniaturization of array spots down to 0.5 mm.
