In Vivo Subcellular Mass Spectrometry Enables Proteo-Metabolomic Single-Cell Systems Biology in a Chordate Embryo Developing to a Normally Behaving Tadpole (X. laevis)**

Camille Lombard-Banek et al.

Summary: The study reports the development of in vivo subcellular high-resolution mass spectrometry for proteo-metabolomic molecular systems biology in complex tissues. Through precision-translated fabricated microcapillaries, subcellular content of different cell types was analyzed with differential quantitative activities of proteins and metabolites identified. The technology preserved integrity of analyzed cells, neighboring cells, and embryos, with 95% of analyzed embryos developing into sentient tadpoles indistinguishable from wild-type siblings.

Image-guided MALDI mass spectrometry for high-throughput single-organelle characterization

Daniel C. Castro et al.

Summary: This article presents a workflow for image-guided MALDI MS characterization of peptide and lipid content in single organelles, allowing observation of the chemical heterogeneity within and between dense-core vesicles and lucent vesicles.

Altering metabolite distribution at Xenopus cleavage stages affects left-right gene expression asymmetries

Rosemary M. Onjiko et al.

Summary: This study suggests that altering metabolite distribution at cleavage stages in Xenopus embryos can affect early events in L-R patterning, but these effects can be compensated for during organogenesis.

Successive High-Resolution (H2O)n-GC1B and C60-SIMS Imaging Integrates Multi-Omics in Different Cell Types in Breast Cancer Tissue

Hua Tian et al.

Summary: This study developed a new methodology for comprehensive lipidomic and metabolomic profiling of individual cells on frozen-hydrated tissue sections, combined with cell-type specific lanthanide antibodies analysis. The research revealed distinct variations of distribution and intensities of key ions in different types of cells within the tumor microenvironment. The feasibility of employing SIMS imaging for multi-omics profiling at the single-cell level in the same tissue section opens up possibilities for gaining new insights into cellular interactions in various tissue microenvironments.

Metabolomic profiling of single enlarged lysosomes

Hongying Zhu et al.

Summary: SLMS integrates lysosomal patch-clamp recording and induced nanoESI/MS for concurrent metabolic and electrophysiological profiling of individual enlarged lysosomes. This technique has been validated for accuracy and reliability, enabling the classification of lysosomes into subpopulations based on their chemical and biological differences in various cell types. SLMS has the potential to investigate heterogeneous lysosomal metabolic changes during physiological and pathological processes.

SpaceM reveals metabolic states of single cells

Luca Rappez et al.

Summary: SpaceM is an open-source method for in situ single-cell metabolomics that can detect a large number of metabolites per hour and predict and validate the cell types and metabolic states. By simulating different stimuli, SpaceM reveals the coexistence of different cellular metabolic states and the mechanisms of balance disruption.
