The UBA domain of conjugating enzyme Ubc1/Ube2K facilitates assembly of K48/K63-branched ubiquitin chains

Lukas Pluska et al.

Summary: The assembly of specific polymeric ubiquitin chains on target proteins is crucial for regulating cellular processes, but the mechanisms behind selective synthesis of these chains remain unclear. The E2 enzymes Ubc1 and Ube2K interact preferentially with ubiquitin chains linked through lysine 63 (K63), facilitating the selective assembly of K48/K63-branched ubiquitin conjugates. The activity of the UBA domain in these enzymes is linked to cellular proteostasis maintenance.

RING domains act as both substrate and enzyme in a catalytic arrangement to drive self-anchored ubiquitination

Leo Kiss et al.

Summary: Attachment of ubiquitin to proteins is a versatile and abundant posttranslational modification that affects outcomes in various physiological processes. Substrate binding can induce a specific RING topology that enables self-ubiquitination. Studies have provided insights into the chemical mechanism of ubiquitin conjugation and the formation of ubiquitin chains.

Summary: By determining the structure of a covalently trapped E1-E2-ubiquitin thioester mimetic, researchers identified two distinct conformations of the E1-E2-Ub complex which suggest the presence of an affinity switch mechanism facilitating thioester transfer.
