DeepTracer for fast de novo cryo-EM protein structure modeling and special studies on CoV-related complexes

Jonas Pfab et al.

Summary: DeepTracer is an automated deep learning-based method for fast de novo multichain protein complex structure determination from high-resolution cryo-EM maps. It shows improved residue coverage and rmsd values compared to current state-of-the-art methods, and demonstrates competitive accuracy and efficiency in structure modeling. With exceptionally fast computations, DeepTracer allows for tracing a large number of residues in a short amount of time.

Structural basis for the N-degron specificity of ClpS1 from Arabidopsis thaliana

Leehyeon Kim et al.

Summary: The N-degron pathway regulates the half-life of proteins in bacteria and plants by recognizing the N-terminal residue of substrates. A structural analysis of AtClpS1 reveals differences in N-degron binding compared to bacterial ClpS proteins, highlighting the importance of subtle structural variations in determining substrate selectivity in protein degradation pathways.

Ubiquitin ligation to F-box protein targets by SCF-RBR E3-E3 super-assembly

Daniel Horn-Ghetko et al.

Summary: E3 ligases are typically classified by hallmark domains such as RING and RBR. However, many neddylated CRLs and ARIH family E3 ligases form E3-E3 super-assemblies, which account for nearly half of all ubiquitin ligases in humans. By using activity-based chemical probes and cryo-electron microscopy, it has been shown that the E3-E3 mechanism can ubiquitylate diverse substrates, providing insights into the widespread ubiquitylation mechanism.
