Inspired heat shock protein alleviating prodrug enforces immunogenic photodynamic therapy by eliciting pyroptosis

Bioresponsive immune-booster-based prodrug nanogel for cancer immunotherapy

Summary: The combination of chemotherapy and immunotherapy shows great potential in inhibiting tumor growth and improving the immunosuppressive tumor microenvironment. However, drug bioavailability limits the therapeutic effectiveness. In this study, a bioresponsive doxorubicin-based nanogel was developed for tumor-specific co-delivery of drugs. The nanogel exhibited stability, selective drug release, and prolonged survival time. It induced powerful antitumor immune response and damaged glucose metabolism in glycolysis and the tricarboxylic acid cycle. This nanogel provides a potential strategy to overcome the limitations of immunotherapy and offers a paradigm for immunomodulatory nanomedicines.
