Diversity of Adenostemma lavenia, multi-potential herbs, and its kaurenoic acid composition between Japan and Taiwan

Miwa Maeda et al.

Summary: Adenostemma lavenia is a widely distributed traditional herbal medicine in East Asia, with different chemical compositions and diverse biological activities in different regions. Chemical analysis revealed high levels of specific compounds in Japanese A. lavenia that showed potent anti-melanogenic effects, while Taiwanese A. lavenia contained different compounds with various biological properties such as anti-inflammatory and anti-oxidative effects. The presence of thiol moieties on target proteins may be crucial for the biological activities of specific compounds found in A. lavenia.

Melanosome transport and regulation in development and disease

Xiaoyu Tian et al.

Summary: Melanosomes are organelles responsible for synthesizing and organizing melanin to provide color to skin, hair, and eyes. Disorders in melanogenesis and melanosome transport are linked to various pigmentary diseases, with clinical cases shedding light on molecular mechanisms controlling melanosome-related pathways. A better understanding of melanogenesis and melanosome transport is crucial for the development of diagnostic and therapeutic approaches.

Thioxothiazolidin derivative, 4-OST, inhibits melanogenesis by enhancing the specific recruitment of tyrosinase-containing vesicles to lysosome

Kenta Isogawa et al.

Summary: This study investigates a new thioxothiazolidin derivative (4-OST) which strongly inhibits melanogenesis in mouse melanoma B16F10 cells by reducing tyrosinase protein levels and altering the destination of the tyrosinase vesicle from the melanosome to the lysosome. These findings provide insights for further research on the tyrosinase-specific vesicle transport system.

Development of Pigmentation-Regulating Agents by Drug Repositioning

Seo-Mi-Gon Jeong et al.

Summary: Skin color is determined by melanin synthesis and distribution, and issues in this process can result in pigmentation defects. Through research on substances or drugs associated with pigmentation control, certain medications have been identified as potential therapeutics for pigmentation disorders through drug repositioning.

The in vitro and in vivo depigmenting activity of pterostilbene through induction of autophagy in melanocytes and inhibition of UVA-irradiated α-MSH in keratinocytes via Nrf2-mediated antioxidant pathways

You-Cheng Hseu et al.

Summary: Research has shown that Pterostilbene has anti-melanogenic effects by inhibiting melanin production through autophagy induction. Experimental results indicate that Pt can suppress melanin synthesis by activating the Nrf2 antioxidant pathway and downregulating genes related to the CREB-MITF pathway, such as TRP-1/-2 and tyrosinase.

Human amniotic stem cells-derived exosmal miR-181a-5p and miR-199a inhibit melanogenesis and promote melanosome degradation in skin hyperpigmentation, respectively

Xiao-Yu Wang et al.

Summary: Our study demonstrated that human amniotic stem cells and their conditioned medium significantly reduced hyperpigmentation in both in vivo and in vitro settings, by inhibiting melanin synthesis and promoting melanosome degradation. Additionally, exosomes derived from human amniotic stem cells contained microRNAs that suppressed melanogenesis and promoted melanosome degradation, suggesting potential therapeutic applications for skin hyperpigmentation disorders.
