Cyclooxygenase-2 Inhibitor: A Potential Combination Strategy With Immunotherapy in Cancer

Summary: COX-2, frequently expressed in cancer cells, is associated with poor prognosis. PGE2, a key factor in immunosuppression, triggers tumor immune evasion and leads to poor response to immunotherapy. COX-2 inhibitors may help to restrain the immunosuppressive effects of PGE2 and enhance or reverse the response to immune checkpoint inhibitors.

Upregulation of C/EBPα Inhibits Suppressive Activity of Myeloid Cells and Potentiates Antitumor Response in Mice and Patients with Cancer

Summary: This study evaluated the mechanisms of therapeutic upregulation of the transcription factor C/EBP alpha in preventing tumor progression in patients with advanced hepatocellular carcinoma and different mouse tumor models. The results showed that therapeutic C/EBP alpha small activating RNA treatment led to decreased immune-suppressive myeloid cell numbers and potent antitumor responses. The study demonstrated that combining C/EBP alpha activation with T cell-dependent therapies enhanced the antitumor effects in various tumor models.

MTL-CEBPA Combined with Immunotherapy or RFA Enhances Immunological Anti-Tumor Response in Preclinical Models

Summary: Combination treatment with MTL-CEBPA, anti-PD-1 checkpoint inhibitors, and radiofrequency ablation showed promising results in preclinical models by reducing tumor size, modulating the tumor immune microenvironment, and increasing tumor infiltrating lymphocytes, suggesting a potential clinical application in enhancing anti-tumor immunity.
