Optimization of a transient antibody expression platform towards high titer and efficiency

Elizabeth Greene et al.

Summary: This study presents the optimization of parameters for TGE for antibodies from CHO cells, resulting in a uniquely optimized process with higher titer and reduced costs and time. It also investigates the amount of coding DNA used in TGE and the influence of kinetics and size of the transfection complex on the in vitro efficiency of the transfection, presenting the first report of an optimized TGE platform using Filler DNA for the screening and production of therapeutic mAbs in an early drug discovery setting.

A review of formulations of commercially available antibodies

Robert G. Strickley et al.

Summary: This review examines 126 commercially available antibodies approved globally between 1986 and February 2021, including various formulations, routes of administration, and formulation components. The number of approved antibodies per year has increased significantly after 2014. Most antibody products are packaged in single-dose units, with different routes of administration and formulation components.

Highly sensitive detection of antibody nonspecific interactions using flow cytometry

Emily K. Makowski et al.

Summary: The PolySpecificity Particle (PSP) assay is a sensitive flow cytometry method for evaluating antibody nonspecific interactions, overcoming previous limitations and allowing for evaluation of diverse types of antibodies. This assay is more sensitive than standard assays like ELISAs, and has the potential to accelerate the development of safe and effective antibody therapeutics.
