Unique roles of rare variants in the genetics of complex diseases in humans

Yukihide Momozawa et al.

Summary: Rare variants sequencing analysis has revealed unique roles in the genetics of complex diseases in humans, providing hypothesis-free evidence for gene causality, a precise target of functional analysis for understanding disease mechanisms, a new favorable target for drug development, and a genetic marker with high disease risk for personalized medicine. As whole-genome sequencing continues to identify more rare variants, the roles associated with rare variants will also increase. However, a better estimation of the functional impact of rare variants across the whole genome is needed to enhance their contribution to improvements in human health.

Contribution of homozygous and compound heterozygous missense mutations in VWA2 to Alzheimer's disease

Julie Hoogmartens et al.

Summary: Alzheimer's disease is the most frequent diagnosis of neurodegenerative dementia, with early and late onset categories, affecting both familial and sporadic patients. A study identified that homozygous and compound heterozygous missense mutations in the VWA2 gene may contribute to the risk of Alzheimer's disease in sporadic patients.

Summary: This study identified two novel genetic variants associated with AD, highlighting the importance of APOE epsilon 4 non-carriers and providing new insights into the pathology of AD. The findings suggest that age-of-onset of AD shows a stronger genetic signal than AD case-control status, capturing many new variants and revealing potential biological functions.

Opportunities and challenges for the computational interpretation of rare variation in clinically important genes

Gregory McInnes et al.

Summary: Genome sequencing enables precision medicine by tailoring treatment to individual genetic variations. Understanding the function and clinical impacts of rare genetic variants is crucial, particularly in genes of known importance. A genomic learning healthcare system is proposed to continuously collect and assess rare variants, with a focus on early-onset treatable diseases in newborns and actionable pharmacogenomics, utilizing emerging machine learning methods for prediction and considering ethical implications.

A genome-wide association study with 1,126,563 individuals identifies new risk loci for Alzheimer's disease

Summary: This study identified seven new genetic loci associated with Alzheimer's disease through a genome-wide association study, implicating microglia and immune cells in late-onset disease. The research highlights the relevance of microglia, immune cells, and protein catabolism to late-onset Alzheimer's disease, while prioritizing previously unidentified genes for further investigation. The results are expected to contribute to larger meta-analyses to uncover additional genetic variants affecting Alzheimer's pathology.

Probability of Alzheimer's disease based on common and rare genetic variants

Valentina Escott-Price et al.

Summary: This study developed a method to estimate Alzheimer's disease risk by combining polygenic risk score (PRS), APOE, rare variants, and disease prevalence in age groups. The results showed that AD probability varies with age, APOE status, and presence of rare variants. This approach has the potential for clinical use and can be updated with new rare variants and genome-wide association data.
