Caught in vicious circles: a perspective on dynamic feed-forward loops driving oxidative stress in schizophrenia

Michel Cuenod et al.

Summary: There is a pathological link between oxidative stress and schizophrenia, with oxidative stress playing a central role in the impairment of neural circuits and subsequent cognitive, emotional, social, and sensory deficits characteristic of the disorder. Redox dysregulation, NMDA receptor hypofunction, neuroinflammation, and mitochondria dysfunction exacerbate this damage, creating a vicious cycle that leads to persistent impairments in neural circuit synchronization. Therapeutic approaches targeting these cycles show promise for interventions, and a reverse translation of circuitry approaches is needed for improved early detection and successful clinical trials in precision medicine for psychiatry.

Elevated activity of superoxide dismutase in male late-life schizophrenia and its correlation with clinical symptoms and cognitive deficits

Lijuan Huo et al.

Summary: This study investigated changes in SOD activity and its relationship with psychotic symptoms and cognitive deficits in late-life schizophrenia (LLS). The results showed that LLS patients had higher total levels of SOD compared to controls and there were correlations between SOD levels and psychotic symptoms as well as cognitive performance.

Oxidative Stress-Related Mechanisms in Schizophrenia Pathogenesis and New Treatment Perspectives

Evgeny A. Ermakov et al.

Summary: Schizophrenia is a highly heterogeneous disease at various levels, with oxidative stress-related mechanisms playing a significant role in its manifestation and development, involving genetic causes, mitochondrial dysfunction, metabolic abnormalities, and more. These mechanisms ultimately contribute to the occurrence and progression of schizophrenia at the whole-organism level.

The Nrf2 pathway in psychiatric disorders: pathophysiological role and potential targeting

Ranjana Bhandari et al.

Summary: Psychiatric disorders are linked to oxidative stress, with the Nrf2 pathway playing a crucial role in maintaining balance and interacting with NF-K beta and Nrf2 transcription factor in severe oxidative stress. Various Nrf2 modulators currently in clinical trials show promise in reducing oxidative stress and neuroinflammation, offering potential novel therapeutic options.

The mystery of claustral neural circuits and recent updates on its role in neurodegenerative pathology

Vladimir N. Nikolenko et al.

Summary: This review aims to provide an update on claustrum anatomy, ontogenesis, cytoarchitecture, neural networks and their functional relation to the incidence of neurological diseases. Despite new methods that have made it possible to study the claustrum at the molecular, genetic and epigenetic levels, its functions and connectivity are still poorly understood. Several studies have shown changes in the appearance, structure and volume of the claustrum in neurodegenerative diseases, such as Parkinson's disease (PD), Alzheimer's disease (AD), autism, schizophrenia, and depressive disorders.

Summary: Schizophrenia is associated with increased oxidative stress, potentially leading to a worsening of symptoms and poor outcomes. Targeting oxidative stress may be a promising approach for future pharmacotherapy in schizophrenia.
