Direct Nanopore Sequencing of Individual Full Length tRNA Strands

Summary: This study presents a method for direct tRNA sequencing using the Oxford Nanopore MinION, with custom adapters and an optimized nanopore sequencing pipeline. The method was successfully tested on purified E. coli tRNA samples, demonstrating the potential of nanopore direct tRNA sequencing to detect expected isoacceptors and known modifications. Further improvements are needed for implementing tRNA sequencing for human healthcare applications.
