The Population-Specific Impact of Neandertal Introgression on Human Disease

Michael Dannemann

Summary: Research has found associations between Neandertal-derived DNA and autoimmune diseases, prostate cancer, and type 2 diabetes, with many of these disease associations linked to specific populations' Neandertal DNA.

Massively parallel discovery of human-specific substitutions that alter enhancer activity

Severin Uebbing et al.

Summary: Massively parallel enhancer assays in neural stem cells were used to quantify the functional impact of >32,000 human-specific substitutions in >4,300 human accelerated regions (HARs) and human gain enhancers (HGEs). Over 30% of active HARs and HGEs showed differential activity between human and chimpanzee, with human-specific substitutions interacting in additive and nonadditive ways to modify enhancer function. Substitutions in HARs, which are highly constrained compared to HGEs, had smaller effects on enhancer activity, indicating buffering of the impact of human-specific substitutions in enhancers with constrained ancestral functions.

The cis-regulatory effects of modern human-specific variants

Carly Weiss et al.

Summary: The study utilized massively parallel reporter assays to investigate the regulatory effects of modern human-specific variants, revealing that a portion of these variants drove differential gene expression between human populations, particularly in genes related to vocal tract and brain anatomy and function.

The timing of human adaptation from Neanderthal introgression

Sivan Yair et al.

Summary: The study examines the spatio-temporal history of selection favoring Neanderthal-introgressed alleles in modern human populations, identifying cases where these alleles were quickly beneficial or persisted at low frequency. It also shows that some alleles persisted at low frequency were later independently favored in geographically separated populations. The research highlights how admixture with ancient hominins has contributed to modern human adaptation and contextualizes the observed levels of Neanderthal ancestry in present-day and ancient samples.
