A new reverse transcription recombinase-aided amplification assay was developed to detect oseltamivir-resistant H275Y mutation in a rapid and sensitive manner, providing technical support for precision medicine in individual drug applications for patients with severe infection or immunosuppression.
The emergence of the influenza A(H1N1)pdm09 virus with the NA-H275Y mutation, which confers oseltamivir resistance, must be monitored, especially in patients undergoing neuraminidase inhibitor treatment. In this study, we developed a reverse transcription recombinase-aided amplification assay that has high sensitivity (detection limit: 1.0 x 101 copies/mu L) and specificity for detecting the oseltamivir-resistant H275Y mutation; the assay is performed within 30 min at a constant temperature of 39 degrees Celsius using an isothermal device. This method is suitable for the clinical application of targeted testing, thereby providing technical support for precision medicine in individual drug applications for patients with severe infection or immunosuppression.
