Recent Advances in Software Tools for More Generic and Precise Intact Glycopeptide Analysis

Weiqian Cao et al.

Summary: Intact glycopeptide analysis has been recognized as a key barrier to understanding glycosylation in organisms. MS-based strategies and software tools have greatly facilitated the analysis of intact glycopeptides, with particular focus on intact N-glycopeptides. This review provides a systematic overview of the intact glycopeptide identification process and highlights recent software tools developed for glycopeptide spectrum interpretation and quality control.

Integrated Glycoproteomics Identifies a Role of N-Glycosylation and Galectin-1 on Myogenesis and Muscle Development

Ronnie Blazev et al.

Summary: The covalent addition of glycans to cell surface and secreted proteins plays a crucial role in the development of multicellular organisms. Aberrant protein glycosylation has been linked to muscular diseases, highlighting the importance of glycan-mediated functions in myogenesis. Dynamic remodeling of the membrane-associated proteome during myogenesis was observed, with changes in the abundance of lectins and enzymes facilitating glycan biosynthesis. Myogenesis involves the regulation of glycans and lectins, with key proteins like galectin-1 influencing the differentiation and formation of muscle tissue.

In-depth Site-specific Analysis of N-glycoproteome in Human Cerebrospinal Fluid and Glycosylation Landscape Changes in Alzheimer's Disease

Zhengwei Chen et al.

Summary: A novel N-glycoproteomic approach was developed in this study and successfully applied to analyze human cerebrospinal fluid samples, identifying a large number of intact N-glycopeptides and providing a molecular basis for understanding the structure-function relationships and disease mechanisms related to glycoproteins in cerebrospinal fluid. Additionally, a comparative in-depth N-glycoproteomic analysis of cerebrospinal fluid samples from healthy controls and Alzheimer's disease patients revealed altered glycosylation patterns for several N-glycoproteins, suggesting potential targets for further studies on glycosylation-based Alzheimer's disease research.
