Dynamics of the PriA Helicase at Stalled DNA Replication Forks

Summary: PriA, a DNA helicase, plays a key role in restarting stalled DNA replication forks in bacteria. Through AFM imaging, it was observed that PriA binds to the replication fork and is capable of translocating over distances of about 400 bp. PriA can switch strands during translocation, depending on the type of DNA fork structure.

Heterogeneity in E. coli RecBCD Helicase-DNA Binding and Base Pair Melting

Summary: E. coli RecBCD, a helicase/nuclease involved in double stranded (ds) DNA break repair, binds to a DNA end and melts several base pairs using its binding free energy. The interactions with ssDNA and structural characteristics of RecBCD-DNA complexes were studied, revealing conformational heterogeneities in RecB(Nuc) and RecD. The extent of DNA melting regulates initiation of helicase activity within RecBCD complexes.

SSB Facilitates Fork-Substrate Discrimination by the PriA DNA Helicase

Summary: PriA plays a role in reloading the primosome onto resurrected replication forks, with SSB enhancing its ability to discriminate between fork substrates. The interaction between PriA and SSB increases the catalytic efficiency of the helicase, but also decreases its affinity for other forks. The unique ability of PriA to bind with high affinity to the 3'-OH at the end of the nascent leading strand, in the presence of SSB, ensures the correct loading onto the fork for directed replication restart.
