Membrane-selective nanoscale pores in liposomes by a synthetically evolved peptide: implications for triggered release

Summary: Macrolittins are a novel family of alpha-helical pore-forming peptides that show unprecedented selectivity for releasing macromolecular cargoes from phosphatidylcholine (PC) liposomes over cell plasma membranes. This selectivity is partly explained by the sensitivity of macrolittin activity to physical chemical properties of the bilayer hydrocarbon core. In the presence of cells, macrolittins release vesicle-entrapped cargoes without directly affecting the cells or involving vesicle-vesicle or vesicle-cell interactions.

The analysis indicates a fine-tuned flexibility in membrane permeabilization mechanisms over time and concentration range.
