Organs-on-chips: into the next decade

Summary: OoCs, also known as microphysiological systems or 'tissue chips', have the potential to be informative at multiple stages of drug discovery and development, providing insights into normal human organ function and disease pathophysiology, as well as predicting the safety and efficacy of investigational drugs in humans. The field of OoCs has seen significant advances in recent years, but there are still challenges and opportunities that need to be addressed to fully realize their potential for translational research.

Primary Human Colonic Mucosal Barrier Crosstalk with Super Oxygen-Sensitive Faecalibacterium prausnitzii in Continuous Culture

Summary: This study demonstrates the effects of commensal anaerobe Faecalibacterium prausnitzii on a primary human mucosal barrier using a specifically designed gut microbiome platform. Continuous co-culture with F. prausnitzii in an anaerobic environment leads to increased growth and butyrate production, while also maintaining the stability of the colon epithelial barrier. Additionally, the anti-inflammatory effects of F. prausnitzii are mediated through histone deacetylase (HDAC) and the Toll-like receptor-TLR-nuclear factor kappa B (NF-kappa B) axis, with butyrate playing a significant role in downregulating inflammation-related receptors TLR3 and TLR4.
