Genome mining of Streptomyces sp. CB00271 as a natural high-producer of β-rubromycin and the resulting discovery of β-rubromycin acid

Liwei Yi et al.

Summary: By utilizing a genome mining strategy, a high-producing strain of beta-RUB was identified, leading to the discovery of a new congener beta-RUB acid with improved solubility and higher anticancer activity. This work provides a foundation for the pilot-scale production of beta-RUB and its congeners, as well as offers a practical strategy for exploring other microbial natural products.

Dual-purpose isocyanides produced by Aspergillus fumigatus contribute to cellular copper sufficiency and exhibit antimicrobial activity

Nicholas Raffa et al.

Summary: Fungi maintain the balance of metal micronutrient pools through various homeostasis mechanisms. Metabolites derived from the xan biosynthetic gene cluster in Aspergillus fumigatus bind copper, affect cellular copper content, and exhibit significant metal-dependent antimicrobial properties. These metabolites also demonstrate copper-dependent antimicrobial properties against a variety of human pathogenic fungi and bacteria.

New-to-nature chemistry from old protein machinery: carbene and nitrene transferases

Zhen Liu et al.

Summary: Hemoprotein-catalyzed carbene and nitrene transformations are powerful tools for constructing complex molecules, showcasing the emergence and evolution of new protein catalysts. These laboratory-invented enzymes harness the ability of proteins to tame highly reactive carbene and nitrene species with high selectivity. Recent studies have shown that biocatalysts outperform small-molecule catalysts in terms of selectivity and turnover rates.
