Metal-Organic Frameworks for Photodynamic Therapy: Emerging Synergistic Cancer Therapy

Tracking Reactions of Asymmetric Organo-Osmium Transfer Hydrogenation Catalysts in Cancer Cells

Summary: The cellular stability of a metallodrug complex catalyzing the reduction of pyruvate to lactate in cells was investigated, with the ligand being displaced by endogenous thiols and translocated to the nucleus while the metal fragment is exported from the cells. The efficiency of catalysis is low due to this phenomenon, suggesting that it could be optimized by developing thiol resistant analogues. This work also provides a new way for the delivery of ligands which are inactive when administered on their own.

DNA-Intercalative Platinum Anticancer Complexes Photoactivated by Visible Light

Summary: The photoactivatable complexes formed by combining photoactivatable agents with 1,8-naphthalimides showed enhanced photo-oxidation and cytotoxicity in human cancer cells, with low dark cytotoxicity. Complexes also demonstrated pre-intercalation into DNA, resulting in enhanced photo-induced DNA crosslinking.

Elemental mapping of half-sandwich azopyridine osmium arene complexes in cancer cells

Summary: By studying the accumulation and localization of metal and ligands in cancer cells treated with potent anticancer complexes, it is found that iodido complexes are rapidly activated in cancer cells and the bond between osmium and the chelated ligand appears to remain stable in cells for about 24 hours, although some release of the ligand is observed. These complexes seem to degrade more rapidly in normal human cells, possibly providing a mechanism for selective cytotoxicity towards cancer cells.
