Adult GAMT deficiency: A literature review and report of two siblings

Summary: This study describes two adult cases of GAMT deficiency from a consanguineous family in Pakistan, highlighting the significant intellectual and neurological impairments, as well as contractures and behavioral abnormalities observed in the patients. A homozygous nonsense variant in the GAMT gene was identified through exome sequencing, and the study emphasizes the rapidly progressive nature of musculoskeletal phenotype and neurologic decline compared to previously reported adult cases.
