Delayed inhibition of ERK and p38 attenuates neuropathic pain without affecting motor function recovery after peripheral nerve injury

Saisai Huang et al.

Summary: The study identified the time window for therapeutic interventions by characterizing neuropathic pain and motor function recovery in a mouse sciatic nerve crush injury model. It was found that delayed delivery of a combination of ERK and p38 inhibitors at three weeks post-PNI could significantly alleviate PNI-induced neuropathic pain without affecting motor function recovery. Additionally, the combined use of these inhibitors could suppress pain more effectively than using either inhibitor alone, potentially reducing side effects and risks.

Neuroimmune interactions and immunoengineering strategies in peripheral nerve repair

Kathryn L. Wofford et al.

Summary: Peripheral nerve injuries disrupt cellular communication between the central nervous system and somatic distal end targets, leading to limited muscle reinnervation and functional recovery. Macrophages play a critical role in the regeneration of the injured peripheral nervous system, and implementing macrophage-based immunomodulatory therapies is promising for nerve injuries in various clinical presentations.

Summary: NGF is an important neurotrophin involved in pain signal transmission, and inhibitors have shown efficacy in reducing joint pain in osteoarthritis patients, but higher doses may increase the risk of rapidly progressive osteoarthritis. Ongoing studies aim to better understand the mechanisms involved in this adverse event.

Bi-allelic loss of function variants in COX20 gene cause autosomal recessive sensory neuronopathy

Hai-Lin Dong et al.

Summary: This study identified an autosomal recessive sensory neuronopathy linked to pathogenic variants in the COX20 gene, which plays a vital role in mitochondrial complex IV assembly and is highly expressed in proprioceptive sensory neurons. Loss of COX20 contributes to mitochondrial bioenergetic dysfunction as a mechanism in peripheral sensory neuron disease.
