Resting state functional MRI brain signatures of fast disease progression in amyotrophic lateral sclerosis: a retrospective study

Summary: Advanced neuroimaging techniques have the potential to monitor disease spreading in amyotrophic lateral sclerosis (ALS). Functional and structural magnetic resonance imaging changes were investigated in ALS patients at diagnosis and over 18 months to discriminate fast progressors from slow progressors. Extra-motor network impairment may appear early in ALS patients with faster disease progression, suggesting that more widespread functional connectivity damage may indicate a poorer prognosis.

Specifically, increases in UPDRS scores were linked to cognitive slowing and reduced intra-subcortical network connectivity over time.
