Cell-Type-Specific Impact of Glucocorticoid Receptor Activation on the Developing Brain: A Cerebral Organoid Study

Cristiana Cruceanu et al.

Summary: Objective: Maintaining a balanced activation of glucocorticoid receptor (GR) is crucial for organ formation. Excessive glucocorticoid exposure during pregnancy has been associated with negative outcomes in brain development, but the underlying mechanisms remain unclear. In this study, an in vitro model using induced human pluripotent stem cell (hiPSC)-derived cerebral organoids was used to investigate the impact of prenatal glucocorticoid exposure on the developing brain. The results showed that GR was expressed in all cell types and its activation affected various pathways, including differentiation and maturation, in neurons and progenitor cells. Neurons exhibited differential gene expression associated with behavior-related phenotypes and disorders. This human neuronal glucocorticoid response profile was validated in cerebral organoids from different hiPSC lines. These findings suggest that excessive glucocorticoid exposure in utero could interfere with neuronal maturation and increase susceptibility to neurodevelopmental disorders. Cerebral organoids have great potential for studying the effects of glucocorticoids on early human brain development.

An emerging role for microglia in stress-effects on memory

Jeniffer Sanguino-Gomez et al.

Summary: Stressful experiences can lead to increased levels of noradrenaline and glucocorticoid hormones in the brain, affecting the interaction between microglia and neurons, impairing neuronal function and learning memory, and triggering immune response-related effects that impair memory and synaptic plasticity.

Hippocampal glucocorticoid target genes associated with enhancement of memory consolidation

Jacobus C. Buurstede et al.

Summary: Glucocorticoids enhance memory consolidation of emotionally arousing events through genomic interactions in the hippocampus, affecting gene expression in both neuronal and non-neuronal cells. This effect is largely independent of training and involves differential DNA-binding of the glucocorticoid receptor and regulation of target genes.

Effects of Long-Term Endogenous Corticosteroid Exposure on Brain Volume and Glial Cells in the AdKO Mouse

Jorge Miguel Amaya et al.

Summary: Chronic exposure to high levels of glucocorticoids can have detrimental effects on health, leading to metabolic abnormalities such as those seen in Cushing's syndrome. MRI studies have shown volumetric changes in the brains of CS patients, both during active disease and in remission. Research in the AdKO mouse model for CS revealed brain region-specific changes, including aberrant myelination and white matter damage.

Summary: This study provides a comprehensive analysis of cell types in the mammalian isocortex and hippocampal formation, revealing a shared neural circuit organization between the two structures. The research also uncovers large-scale variations in cell types along different dimensions within these brain regions.

The glucocorticoid receptor specific modulator CORT108297 reduces brain pathology following status epilepticus

Aynara C. Wulsin et al.

Summary: The treatment with C108297 attenuated corticosterone hypersecretion, decreased the density of hilar ectopic granule cells, and reduced microglial proliferation following status epilepticus. However, it did not prevent mossy cell loss in treated mice. This suggests the potential of glucocorticoid receptor modulators as therapeutic agents for preventing brain pathology after status epilepticus.
