The Rise and Fall of Isotonitazene and Brorphine: Two Recent Stars in the Synthetic Opioid Firmament

Marthe M. Vandeputte et al.

Summary: Synthetic opioids, particularly non-fentanyl-related ones, are rapidly growing in the recreational opioid market. Many of these drugs are pirated from early literature and synthesized for online sale. The life cycle of these opioids is generally short, with only a few drugs reaching significant detections. This review focuses on two non-fentanyl opioids, isotonitazene and brorphine, and their emergence and decline in the market.

Etazene (N,N-diethyl-2-{[(4-ethoxyphenyl)methyl]-1H-benzimidazol-1-yl}-ethan-1-amine (dihydrochloride)): a novel benzimidazole opioid NPS identified in seized material: crystal structure and spectroscopic characterization

Marta Siczek et al.

Summary: This study aimed to present the spectroscopic characteristics and crystal structure of the benzimidazole opioid etazene, which has recently emerged on the illegal drug market in Poland. The compound was analyzed using X-ray crystallography, gas and liquid chromatography combined with mass spectrometry, as well as other spectroscopic techniques. The authors described the identification and chemical characterization of etazene, highlighting its importance for the detection of new psychoactive substances in forensic and clinical laboratories.

A Highly Efficacious Carfentanil Vaccine That Blunts Opioid-Induced Antinociception and Respiratory Depression

Lisa M. Eubanks et al.

Summary: The opioid epidemic remains a dire public health crisis, with synthetic opioids such as fentanyl being highly potent and dangerous. Researchers have developed vaccines targeting synthetic opioids like fentanyl and carfentanil, which have shown protective effects in mice, providing new strategies for preventing drug abuse and overdose.

Synthesis, Chemical Characterization, and μ-Opioid Receptor Activity Assessment of the Emerging Group of Nitazene 2-Benzylbenzimidazole Synthetic Opioids

Marthe M. Vandeputte et al.

Summary: Several 2-benzylbenzimidazole opioids, known as nitazenes, have emerged on the illicit market, with isotonitazene being the most frequently encountered member. These compounds, structurally unrelated to fentanyl, have high potency and efficacy, with some analogues even surpassing fentanyl. The unexpected very high potency of the N-desethylisotonitazene metabolite, exceeding that of isotonitazene itself, suggests potential significant in vivo consequences.

DARK Classics in Chemical Neuroscience: Etonitazene and Related Benzimidazoles

Istvan Ujvary et al.

Summary: Etonitazene and its derivatives, developed in the late 1950s by the Swiss pharmaceutical giant CIBA, are potent analgesics with morphine-like effects. Recently, unscheduled synthetic opioids have emerged on the illicit drug market, leading to fatalities. This class of compounds has inspired research and led to the emergence of new psychoactive substances, impacting chemical neuroscience.

Improvements on a chemically contiguous hapten for a vaccine to address fentanyl-contaminated heroin

Hyeri Park et al.

Summary: Unintentional overdose deaths related to opioids and psychostimulants have increased due to adulteration with fentanyl. Developing antibodies capable of binding to fentanyl-contaminated heroin can effectively block the psychoactive effects of the drug on the central nervous system.

Broadly Neutralizing Synthetic Cannabinoid Vaccines

Mingliang Lin et al.

Summary: The study investigated the impact of hapten design on antibody affinity and cross-reactivity, successfully identifying vaccine groups with submicromolar IC50. Additionally, a synergistic effect was found through an admixture vaccine approach to simultaneously weaken the effects of two drug classes.

Immunopharmacotherapeutic advancements in addressing methamphetamine abuse

Jinny Claire Lee et al.

Summary: Methamphetamine is a global illicit psychostimulant known for substance abuse disorders but lacks approved pharmacotherapies. Traditional treatments using small molecule agonists or antagonists targeting drug-specific receptors in the brain have limitations in efficacy. Immunopharmacotherapeutic advancements present a promising alternative to address methamphetamine abuse by eliminating central nervous system receptor occupation through peripheral sequestration of abused drugs.
