A Drug-Free Therapeutic System for Cancer Therapy by Diselenide-Based Polymers Themselves

Chao Wei et al.

Summary: A novel drug-free therapeutic system utilizing synthetic polymers themselves shows effective and broad-spectrum anticancer activities with high selectivity towards cancer cells, which can be controlled by tuning the selenium contents. Mechanistic investigation reveals that the system can induce cancer cells to express excessive reactive oxygen species, leading to cellular apoptosis.

Pulmonary Delivery of Reactive Oxygen Species/Glutathione-Responsive Paclitaxel Dimeric Nanoparticles Improved Therapeutic Indices against Metastatic Lung Cancer

Xidong Tian et al.

Summary: This study introduced redox-responsive PTX dimeric nanoparticles for pulmonary delivery to achieve effective therapeutic outcomes against metastatic lung cancer with reduced systemic and local toxicities. These nanoparticles exhibited optimal drug release behavior, excellent colloidal stability, cellular uptake efficiency, and discriminating cytotoxicity between cancer and healthy cells. In a mouse model of metastatic lung cancer, intratracheally delivered PTX dimeric nanoparticles showed stronger anticancer potential with reduced systemic toxicity compared to traditional Taxol intravenous injection.

Disrupting the Redox Balance with a Diselenide Drug Delivery System: Synergistic or Antagonistic?

Yeon Su Choi et al.

Summary: This study investigates the release of therapeutics in the cytosol using a diselenide-based drug carrier, identifying GSH as the major trigger for diselenide cleavage and demonstrating synergistic anticancer effects when combining the carrier with an anticancer drug. The research shows that the combined use of diselenide-based drug carrier and drug has a significant inhibitory effect on tumor growth in vivo.
