Polyubiquitin gene Ubb is required for upregulation of Piwi protein level during mouse testis development

Bitnara Han et al.

Summary: Analysis of the testis proteome in Ubb knockout mice revealed 564 proteins significantly differentially expressed, with 36 downregulated proteins involved in various stages of spermatogenesis. Functional gene ontology analysis identified decreased levels of piRNA metabolic process-related proteins, such as Piwil2 and Tdrd1, in Ubb-null testes. Protein-protein interaction mapping showed that 24 testis development-related proteins, including Hsp90aa1, Eef1a1, and Pabpc1, were directly impacted by the depletion of ubiquitin, with mRNA levels of these proteins reduced in Ubb-knockout testes resembling the global downregulation of piRNA-metabolic gene expression at transcriptional and post-transcriptional levels.

Multiple transcriptomic profiling: p53 signaling pathway is involved in DEHP-induced prepubertal testicular injury via promoting cell apoptosis and inhibiting cell proliferation of Leydig cells

Junke Wang et al.

Summary: The research revealed that DEHP disrupts testicular development, lowers serum testosterone levels in prepubertal mice, induces cell apoptosis and cell cycle arrest in TM3 cells. The p53 signaling pathway plays a crucial role in DEHP-induced prepubertal testicular injuries by promoting cell apoptosis and inhibiting cell proliferation.

Summary: The study found that prenatal smoking was associated with significantly increased weight-adjusted AGD in female infants, with a correlation between mothers' daily smoking rates and AGD. However, fetal smoke exposure in male infants did not show any association with AGD measurements, SPL, and penile girth.

Increased m6A modification of RNA methylation related to the inhibition of demethylase FTO contributes to MEHP-induced Leydig cell injury

Tianxin Zhao et al.

Summary: DEHP, a common environmental endocrine disrupting chemical, induces testicular injury through modulating m6A modification. The study reveals that FTO-mediated m6A modification plays a critical role in Leydig cell injury induced by DEHP.

Decabromodiphenyl ether (BDE-209) exposure to lactating mice perturbs steroidogenesis and spermatogenesis in adult male offspring

Debarshi Sarkar et al.

Summary: The study showed that maternal exposure to BDE-209 during lactation significantly affects male reproductive health in adult offspring, leading to altered testicular histoarchitecture, decreased testosterone production, down-regulation of certain steroidogenic markers, as well as interference with germ cell dynamics and oxidative status in testes.

Occurrence and spatial distribution of legacy and novel brominated flame retardants in seawater and sediment of the South China sea

Hongru Feng et al.

Summary: The occurrence and spatial distribution of PBDEs and NBFRs in the South China Sea in 2018 were investigated, with BDE-209 and decabromodiphenyl ethane identified as predominant BFRs. Rarely detected NBFRs, tris(2,3-dibromopropyl)isocyanurate and 2,4,6-tris(2,4,6-tribromophenoxy)-1,3,5-triazine, were found for the first time in the study area, indicating their widespread application in the Pearl River Delta. Preliminary risk assessment suggests low to medium risks to marine organisms at some sites.

New insights into the mechanisms underlying 5-fluorouracil-induced intestinal toxicity based on transcriptomic and metabolomic responses in human intestinal organoids

Daniela Rodrigues et al.

Summary: This study utilized 3D human colon and small intestine organoid models to investigate the transcriptomic and metabolomic responses to 5-FU. The results revealed that 5-FU impacted molecular pathways such as cell cycle, apoptosis, and mitochondrial ATP synthesis, as well as induced tissue-specific metabolic alterations. Integration of multi-omics identified the transcription factor E2F1 as a key regulator in the toxicity mechanisms of 5-FU.

Long-term exposure to environmental levels of phenanthrene disrupts spermatogenesis in male mice

Jie Huang et al.

Summary: The exposure to Phe in male mice led to accumulation of Phe in the testis, decreased number of spermatogonia and cells, increased apoptosis, and inhibited the function of Sertoli and Leydig cells. Additionally, it upregulated estrogen receptor alpha and downregulated androgen receptor expression, indicating impaired spermatogenesis and potential reproductive toxicity in mammals.

Persistent organic pollutants and couple fecundability: a systematic review

Linda G. Kahn et al.

Summary: The review synthesized evidence on the associations of four common groups of POPs with couple fecundability, with the strongest evidence found for adverse associations of female exposure to polychlorinated biphenyls with time to pregnancy and some additional support for associations of female exposure to polybrominated diphenyl ethers and PFAS with longer time to pregnancy. However, the review provided little or no support for associations between female exposure to OC pesticides or male exposure to any of the POP groups and time to pregnancy.
