Synthesis and characterization of thermosensitive hydrogel based on quaternized chitosan for intranasal delivery of insulin

Summary: The study focused on developing a novel formulation for intranasal insulin delivery using chitosan, HTCC, and Gel, which demonstrated low gelation time, uniform pore structure, and desirable swelling rate for adequate encapsulation and prolonged release of insulin within 24 hours. The optimal samples released 65% of the total amount of insulin in the first 24 hours, showing favorable results for the study.

Despite limitations, an experimental protocol for aerosolized intranasal delivery is presented for better targeting the olfactory epithelium.

Intranasal vasopressin modulates resting state brain activity across multiple neural systems: Evidence from a brain imaging machine learning study

Summary: Arginine vasopressin (AVP), a neuropeptide with widespread receptors in brain regions important for socioemotional processing, is critical in regulating various mammalian social behavior and emotion. The study demonstrated that the role of AVP in socioemotional functioning recruits multiple brain networks distributed across the whole brain rather than specific localized neural pathways. This data-driven approach also opens a novel avenue to delineate neural underpinnings of various neuropeptides or hormones.

Summary: Intranasal drug administration is a common route for therapeutic formulations, but challenges such as poor absorption and mucosal tissue damage exist. Potential absorption enhancers like alkylsaccharides have been investigated to address these issues, improving absorption of therapeutic agents across mucosal membranes and applicable to a wide range of molecules. Alkylsaccharide excipients, such as dodecyl maltoside, have shown to be nonirritating and effective in enhancing systemic bioavailability, and are used in FDA-approved intranasal formulations for migraine and epilepsy patients.

Summary: Efforts were made to develop an effective and biosafe intranasal formulation for parathyroid hormone (1-34), with 10% Kolliphor(R) HS.15 identified as a permeation enhancer that significantly increased the bioavailability of PTH(1-34). Further research on intranasal formulations with higher concentrations of Kolliphor(R) HS.15 for enhanced bioavailability of PTH(1-34) could be considered.
